1.Ullistat acetate is a new oral emergency contraceptive, which is the active chemical component of Ella, a new generation of emergency contraceptive currently marketed in the United States. It can be administered for up to 120 hours after a woman has unprotected sex, and the efficacy of emergency contraception does not decrease with time delay. It is also safe and well tolerated. Ullistat acetate has a wider clinical applicability than levonorgestrel, the most commonly used emergency contraceptive, and has the potential to prevent more unintended pregnancies. Ullistat acetate was developed by HRA Pharmaceuticals and approved by the US Food and Drug Administration in August 2010 under the trade name Ella. For the prevention of pregnancy within 120h after unprotected sex or known or suspected contraceptive failure. Ullistat acetate is a selective progesterone receptor modulator that acts as emergency contraception mainly by inhibiting ovulation. Ullistat acetate was found to inhibit discharge more effectively than levonorgestrel when administered closer to discharge day (when the chance of conception is highest), suggesting that Ullistat acetate may be more effective as an emergency contraceptive.
2.Ulipristalacetate is a progesterone agonist/antagonist whose mechanism of action is inhibition or delay of ovulation; However, changes in the lining of the uterus may also contribute to the benefit. Ullistat acetate is a selective regulator of progesterone receptors and has antagonistic and partial excitatory effects on progesterone receptors. It binds to the progesterone receptor in the body and prevents progesterone from binding to the progesterone receptor. The effectiveness of Ullistat acetate depends on when the drug is administered during the menstrual cycle. Administration during the metaphase of follicular hyperplasia inhibited follicular production and decreased estradiol concentration. Administration during the peak of luteinizing-hormone can delay follicle rupture for 5 to 9 days. Early luteal administration did not significantly delay endometrial maturation, but reduced endometrial thickness by 0.6±2.2mm (mean ±SD).
